Holmium-loaded AcAc nanospheres are potential agents for radionuclides endoradiotherapy because of their low density, biodegradability and favorable radiation characteristics. In this study, production, quality control and biodistribution studies of 166Ho-AcAc have been presented. Firstly, Stable 165Ho acetyl acetone (AcAc) spheres were prepared by the solvent evaporation technique. 165Ho-AcAc Nanospheres converted to 166Ho-AcAc by irradiation in the research reactor at the Tehran. Morphology and stability of the complex were investigated by SEM and ITLC respectively. The complex solutions (100 μCi/100 μl) were injected intra-tail into rat followed by scarification studies post injection. Results indicated that 166Ho-AcAc was prepared successfully with the optimal mean particle size 70-100 nm and displayed a smooth surface with high radiochemical purity more than 97%. The complex was stable at 4ºC, human serum, PBS buffer and room temperature. The highest %ID/g of 166Ho-AcAc was observed in the liver. Our data showed that AcAc nanospheres can be made in the optimal size range for later irradiation and their ability to retain 166Ho makes them attractive agents for undo radiotherapy.
